Orcein as a mucin stain for gastrointestinal tissue sections.

چکیده
Although orcein has been in use for a long time as a stain for elastic fibres, it is only recently that it has been rediscovered for the demonstration of a variety of other substances. These include hepatitis B surface antigen (HBsAg),' 2 copper-associated protein,34 and more recently, intestinal sulphated mucins.35 Sipponen5 studied the normal distribution of orceinpositive mucin in the gastrointestinal tract and compared the results with those obtained by other mucin stains. We here report our findings using a slightly modified orcein staining method after oxidation, on a variety of gastrointestinal lesions as well as normal gastrointestinal tissues.

Methods for detection of hepatitis B surface antigen in paraffin sections of liver: a guideline for their use.
Methods for the localisation of hepatitis B surface antigen (HBsAg) in paraffin sections of the liver include the detection of ground-glass hepatocytes and the use of Shikata's orcein stain, and of immunoperoxidase and immunofluorescent techniques. A comparative study of the different methods on 20 livers shows the orcein stain to be the method of choice for routine use. The Shikata stain is no...
متن کاملThe selective affinity of anionic detergents for elastica: a new stain for elastica.
This report concerns the observation that certain anionic detergents have a specific affinity for elastica, and because these anionic detergents precipitate cationic basic dyes in aqueous solution, a rather specific stain for elastica can be obtained. There are many satisfactory methods for the histologic demonstration of elastica, the most popular being: a) acid orcein (Unna), b) Verhoef's, c)...
متن کاملPresence of perivenular elastic fibers in nonalcoholic steatohepatitis Fibrosis Stage III.
Elastic fibers appear in extensive old fibrotic foci in general. We examined an association between hepatic fibrosis stage and the presence of perivenular elastic fibers in nonalcoholic steatohepatitis (NASH). A total of 48 liver needle biopsy specimens were used, taken from 48 cases with NASH. Fibrosis Stage (Brunt E, et al. Am. J. Gastroenterol. 1999) of the cases was as follows; six Fibrosis...
متن کاملOrcein-alcian blue staining: a new technique for demonstrating acid mucins in gastrointestinal epithelium.
Orcein-alcian blue staining, a new method for the simultaneous demonstration of sulphated and sialomucins in gastrointestinal epithelium was compared with the standard high iron diamine-alcian blue technique. Sections were oxidised with potassium permanganate and decolourised in oxalic acid. They were stained with orcein for four hours, differentiated for a few seconds in acid alcohol, and then...
متن کاملIonic and non-ionic bonds in staining, with special reference to the action of urea and sodium chloride on the staining of elastic fibres and glycogen By D. J. GOLDSTEIN
(a) by competing for hydrogen bonding sites in the tissue or on the dye particle, it may inhibit staining in which the dye-substrate link is a hydrogen bond; (6) because it is a dipole, urea may have some affinity for charged sites in the tissue, and thus tend to reduce electrostatic staining; (c) urea may in some cases decrease staining by increasing the solubility of the dye; (d) by inhibitin...
